University of Leiden , Netherlands open position for PhD Thesis :
Research team of Prof. JP. Abrahams has developed new technologies for studying the structure of proteins by electron diffraction of crystals that are smaller than one micrometer in size. Such crystals are beyond the reach of even the most powerful synchrotron X-ray radiation. In this project there is a vacancy for a Ph. D. with a Masters degree in Chemistry or Physics with an interest in scientific computing and/or electron microscopy/diffraction and/or (protein) X-ray crystallography. We propose to build on our current progress and determine the structure of a protein using electron diffraction patterns of sub-micron 3D crystals. The project is divided in several sub-projects:
Optimize sample handling procedures and/or electron diffraction protocols (collaboration with TU Delft);
Optimize data collection using new quantum area detectors (collaboration with CERN);
Improve the robustness of the crystal indexing procedure;
Adapt computational procedures from X-ray crystallography for integrating and merging the 3D data;
in first instance, the integrated data will be phased by molecular replacement.
The Ph. D. student will work on one or more of the above sub-projects, depending on previous experience and interest of the candidate.
